Storage
This product is stable for only 3 weeks after receipt when handled aseptically and stored at 2-8°C (Do not freeze). After prolonged storage, some precipitate may be observed. This is normal for this product.
Clarify out the aggregates by spinning in centrifuge tube for 2 minutes.
Biological Activity
Sample lots of Oxidized LDL are evaluated for receptor binding to peritoneal macrophages in conjunction with our DiI-Ox-LDL and [I-125] Ox-LDL.
Preparation
Human LDL (which was purified to homogeneity via ultra centrifugation (1.019-1.063g/cc)) is extensively oxidized with Cu2SO4 (oxidant) in PBS at 37°C. Oxidation is terminated by adding excess EDTA-Na2. Each lot is analyzed on agarose gel electrophoresis for migration versus LDL.
This lot of Ox-LDL migrates 3.0fold further than the native LDL. This product can produce potent oxidative stress and can be used to induce cell apoptosis/death (>50%) and produce cell injure model.
TBARS
TBARS is determined colorimetrically by using Malondialdehyde as a standard.
